The mathematical model of the bacterial PEP-dependent phosphotransferase sysem

The detailed kinetic model of the bacterial PEP-dependent phosphotransferase system carrying out transport of the glucose into the cell with concomitant phosphorylation was constructed. The model includes description of the operation of the enzymes EI, HPr, EIIA, EIICB and consists of the differential equations for the concentrations of the different states of the enzymes and substrates. The kinetic parameters of the model (rate constants and equilibrium constants for the elementary stages) were defined by the fitting of the theoretical dependencies to the experimental data on the analogous dependencies. Model describes satisfactorily the experimental data on the distribution of the system’s components after reaching equilibrium, on the steady-state rates of the work of the enzymes and on the kinetic of the phosphorylation of the enzymes and glucose. We used both data for the separate enzymes and for the total system. The temperature and pH influence on the enzymes operation was also described. We also analyzed the behaviour of the system with different substrate concentrations.
