Potassium phenozan as an adaptogen to stre3ss conditions

Mitochondria being the energy exchange regulating site play an important role in the organism response to stress conditions that result in an increase in the level of reactive oxygen species (ROS) in cells (V.A. Baraboi, 1991). The main body of ROS is formed in mitochondria upon an incomplete one- and two-electron reduction of oxygen to superoxide (O2•) or to hydrogen peroxide, respectively (V.P. Skulachev, 2001).

Supposedly, the principal property of adaptogens is a decrease in the excessive production of ROS that results in the activation of lipids peroxidation (LP) in biological membranes. For the purpose of studying the antistress properties of the preparations, a mitochondria “aging” (long-term storage) model was constructed. A test subject used was potassium phenozan (a potassium salt of 2,6-di-tert-butyl-4-hydroxyphenyl-propionic acid) that was introduced to a mitochondria incubation medium.

“Aging” resulted in enhancing the fluorescence intensity of LP products by a factor of 3 to 4. The addition of the preparation resulted in decreasing the LP products fluorescence, which was dose-dependent. The preparation concentration 10-5, 10-6, and 10-7 M produced an insignificant effect on the intensity of LP processes. The potassium phenozan concentrations 10-8- 10-16 and 10-18 - 10-22 M resulted in decreasing the fluorescence intensity of POL products to a control level.

The introduction of the preparation to rats at a dose of 10-14 M resulted in increasing the content of unsaturated fatty acids and decreasing the content of saturated fatty acids in mitochondria membranes 1 h after the injection. As a result, the ratio of unsaturated acids to saturated acids increased from 1.27±0.10 top 2.04±0.06. The changes affected the physiological indices. The introduction of the preparation to animals at the above dose enhanced by 20 to 30% the survival rate of animals under conditions of hypoxia and a low temperature stress. Thus, in terms of the mitochondria “aging” model it is possible to carry out a screening of preparations for antistress properties.
