Concentration optimum plant growth regulator - melaphen when exposed to the membrane of animals

Growth regulator of plants - melafen (a melamine salt of bis (hydroxymethyl) phosphinic acid) was tested as a biologically active substance on the objects of animal origin. The main purpose was to determine the melaphen concentrations not damaging animal cells. Therefore, studies were conducted in a wide range of concentrations (10-21 M-10-2 M). It is known that for pretreatment of crop seeds used aqueous solutions at a concentration melaphen 10-10,

9-11M. The yield of crucifers and cereals increased by 11% or more due to increased stress tolerance of plants under adverse conditions. Increasing the concentration melaphen to 10-8, 10-7 M inhibits the growth of plants. In case of contact melaphen to animal’s first target, to which it can act, are the cell membranes, their protein and lipid components, as well as soluble proteins and blood cells properly. In this study it was shown that the lipid microdomains in a model system multilayer liposomes (produced from individual phospholipid liposomes) change their organization in the presence of melaphen (10-14-10-8 M), as evidenced by changes in the parameters of thermally induced phase transitions. However, in the model is more approximate to the natural membrane, multilayer liposomes from egg lecithin melafen (10-21-10-6 M) did not induce the broadening of the baitlayers and does not alter the distance between the liposomes in multiple associations, according to small-angle X-ray diffraction. In the shadows of red blood cells, a model protein-lipid membrane, melafen even at high concentrations changes in the organization of protein-lipid microdomains does not cause. Soluble blood protein - bovine serum albumin alters the conformation in the presence of melaphen (according to the registration of the intrinsic fluorescence): high melaphen concentrations lead the fiber, splitting the medium and small concentrations leads compaction of the molecule.

At the level of whole cells by light scattering, reported about inhibition of Ca2 +-signal (10-8 M and more) and, by immunoblotting, - about activation of apoptosis (10-10 M) in EAC cells. On erythrocytes in vitro by atomic force microscopy was shows the morphological changes at different melafen concentrations (10-12 M 10-6, 10-4 M).

The main conclusion: when using melaphen as plants growth regulator at concentrations of 10-10, 10-9 M should be treated with caution, because contact of this substance in concentrations higher than 10-13 M with objects of animal origin leads to inhibition of some important cellular processes.
