The features of action of biologically active substances over a wide range of concentrations on the cell targets

For explorations some biologically active substances (BAS) were selected: hydrophilic melafen, synthesized in IOFH RAS for promotion of plant growth, and hydrophobe phenozan, received in joint project of Moscow petroleum processing plant and Institute of Chemical Physics RAS. Phenozan was wounded for stabilization of polymers. Later in IBHF RAS for the solving of biological missions have been synthesized some derivatives of phenozan: amphiphilic phenoksan - potassium salt of phenozan-acid [β-4-oxi-(3, 5-ditertbutyl-4-oxiphenyl) potassium propionate] and hydrophobic hybrid antioxidants IHFANs – alkyl-dimethyl-[β-(4-hydroxi-3,5-ditertbutylphenyl) propionylethyl] ammoniahalogenides). Melafen (melamine’s derivative of bis phosphinic acid) - plant growth regulator. Because this preparation used for the seed treatment with wide distribution it was necessary to investigate its action at the cellular level in the body of warm-blooded animals. The same missions had been delivered and for phenozan and its derivatives.

When any BASs introduce to the body of warm-blooded animal in any manners and any ways, the first immediate targets for BAS are the membrane structures and blood components. These is why experimental objects were 1) model membranes - the multilayer liposomes, simulating the multilayer cellular structures; 2) erythrocyte ghosts with identified by the protein composition, reflecting the cytoskeleton structure of most cells of animal body; 3) soluble blood protein - bovine serum albumin; 4) erythrocytes and others cells.

The studies of the effects of BAS on structures of experimental objects were held by the DSC method, method of small-angle X-ray scattering, EPR and AFM. The tests of the BAS influences to the functional activities of bio-objects were made by potentiometric methods and with using of primary light diffusion at a right angle. It has been ascertained that BAS in wide concentration range (10-17 М - 10-3 М) caused the logged changes of structure and functions of objects.

The comparing of obtained polymodal dependences with famous literature data about some parameters of physical-chemical properties of emulsions IHFAN-10 indicate the change of value of reacting BAS-doses in biologic objects. There were discovered the formation of own phase of IHFAN-10 in membranes. That indicates that the application of hydrophobic BAS need of low and ultra small doses using. Structural change of membranes by hydrophilic melafen and fenoksan alludes to the fact that the mode of action on biologic objects is mediated by the water medium. It is under discussion that the BAS influences may be mediated by probability of existence of supramolecular structures, formed by low doses of BAS in solution and waters molecules. It was the small probability of their existence in the cell. Alternatives to such version are action of BAS to the composition of dissolved gases in vivo and to the attendance of gas bubbles in solution in vitro in solutions.
